La Orquesta se formó a comienzos de 1937 en La Habana por integrantes de la Orquesta de los Hermanos Castro, y optó por el nombre Orquesta Casino de la Playa porque un night club homónimo les ofreció contrato permanente si lo adoptaban. Los miembros principales eran Guillermo Portela (violín), Anselmo Sacasas (piano) y Miguelito Valdés (voz). Después, los siguieron casi todos los integrantes de los Castro, exceptuando a los propios hermanos. En el verano de ese año, el sello Víctor realizó grabaciones en La Habana, en la que participaron 24 agrupaciones de diferente formato. La penúltima fue la recién fundada Casino de la Playa, con seis números, entre los que se encontraba Bruca Maniguá, la célebre canción afro de Arsenio Rodríguez. La banda estaba formada por Guillermo Portela, violín; Luduvino Pereira, Alfredo Saenz y Rodríguez, maderas (saxos y, al parecer, algún clarinete); Walfredo de los Reyes y Luis Rubio, trompetas; Unésimo González, contrabajo; Miguelito Valdés, percusión y voz; Anselmo Sacasas, piano y dirección; Ernesto de la Vega, guitarra y maracas. Siempre se mantuvieron como una agrupación de blancos, con la excepción de Miguelito Valdés, mestizo claro, cuya presencia era aceptada en los exclusivos y excluyentes lugares donde solían tocar. Además de los músicos de plantilla, solían incluir a otros para las grabaciones, como Arsenio Rodríguez (tres) o Ramoncito Castro (bongó), ambos negros y cuyo color no se escuchaba en los discos. En los tres primeros años de su existencia la Orquesta Casino de la Playa realizó un total de 60 grabaciones para la RCA Víctor. En el cuarenta, Sacasas y Miguelito Valdés la abandonaron y se instalaron en los Estados Unidos, donde desarrollaron sus propias carreras. En noviembre de 1945 comienza una nueva etapa en la orquesta, con la inclusión como cantante de Orlando Guerra, más conocido como "Cascarita", y, por presiones de éste, entra un nuevo pianista, que también haría historia: Dámaso Pérez Prado. A partir de 1948, año en que Cascarita abandona a la Casino de la Playa, la orquesta sufre altibajos, hasta desaparecer a finales de los cincuenta o comienzos de los sesenta.
